712 UV-induced reduction in polycomb repression promotes epidermal pigmentation

چکیده

Ultraviolet (UV) radiation is a prime environmental stressor that our epidermis exposed to on daily basis. To avert UV-induced damage, epidermal stem cells (EpSCs) become pigmented via process of heterotypic interaction between melanocytes and EpSCs, however the molecular mechanisms this are not well understood. In study we show function key chromatin regulator, Polycomb complex, was reduced upon UV exposure in human mouse epidermis. Genetic ablation subunits murine mimicking depletion exposure, results an increased number subsequent pigmentation. Genome-wide transcriptional studies regulates expression UV-responsive genes identify type II collagen as critical secreted regulator melanogenesis Altogether, findings how UV-exposure induces Polycomb-mediated changes EpSCs affect melanocyte behavior promote

Knocking off their Sox: lineage-specific repression by Polycomb in epidermal stem cells.

A recent publication in The EMBO Journal (Bardot et al, 2013) provides novel insights into lineage specification during the development of the mouse skin. Ezhkova and colleagues demonstrate that Ezh1 and Ezh2, core enzymes of the Polycomb Repressive Complex (PRC), restrict differentiation of Merkel cells, a specialized population of mechanosensory cells by directly repressing the cell fate dete...

Polycomb Repression under the Skin

Polycomb group proteins control a hierarchy of gene expression during the differentiation of stem cells. In this issue, Ezhkova et al. (2009) show that repression mediated by Polycomb proteins controls the timing of differentiation of precursor cells of the epidermal lineage.

Polycomb CBX7 promotes initiation of heritable repression of genes frequently silenced with cancer-specific DNA hypermethylation.

Epigenetic silencing of genes in association with aberrant promoter DNA hypermethylation has emerged as a significant mechanism in the development of human cancers. Such genes are also often targets of the polycomb group repressive complexes in embryonic cells. The polycomb repressive complex 2 (PRC2) has been best studied in this regard. We now examine a link between PRC1 and cancer-specific g...

Repression of methylcholanthrene-induced epidermal hyperplasia by hydrocortisone.

The present study was undertaken in the light of interest engendered by conflicting reports on the effect of glucocorticoids upon the develop ment of skin tumors in mice treated with polycyclic hydrocarbon carcinogens (2, 4, 7-9, 13, 14, 17, 20, 21). Some investigators also found that systemically administered cortisone did not influ ence the increased mitotic rate (12) or epidermal hyperplasia...
